一六二二年（天啟二年）荷蘭人再次要求明國通商不成，再度進駐澎湖。

占領南部的荷蘭人，是先禮後兵的，先占無人小島「一鯤鯓」，再購地取水飲用之後，發現臺灣人民善良可欺，所謂「人善被人欺，馬善被人騎」，而且沒有武器，又不能團結，各自為政，因此荷蘭人貪念一起，就用武力做為後盾，強占整個臺灣。
一六二四年荷蘭人進入安平一帶的無人小島一鯤鯓，在一鯤鯓修築城堡，命名為奧倫治（Orange），至一六二七年更名為熱蘭遮城（Zeelandia）即今所稱之『安平古堡』，荷蘭總督駐紮於此。（按：當時一鯤鯓與臺灣本島間，尚有一水域，即中國文獻上所稱之「臺江」）
一六二五年荷蘭人以十五匹康甘布（Cangan）向『平埔族人』（按：確實是向西拉雅族人）換取臺江對岸的赤崁（今臺南市中心區），荷蘭人在此取水，並建築倉庫、宿舍，後逐漸發展出一個商業市街，並且在此建普洛文蒂亞城
（Provintia）即今之『赤崁樓』。
